Chemotherapy induced peripheral neuropathy (CIPN)

● Symptoms and Causes  

The symptoms of chemotherapy induced peripheral neuropathy depend on the type of chemotherapy and which nerve fibers are affected.In chemotherapies that affect mainly the sensory nervefibers, the patients experience unusual sensations (paresthesias)nuinbness, balance problems or pain. In cases where the motor nerves are affected, the patients may experience weakness of themuscles in the feet and hands.

Chemotherapy can cause degeneration of peripheral sensory and motor nerves and cause patients to present with sensory disturbances,balance problems or weakness.

Cisplatin Induced Rat CIPN Model

【Mechanism】Cisplatin is the first member of a class of platinum-containing anti-cancer drugs which have their effect by causing crosslinking of DNA, leading to apoptosis. Unfortunately, the platinum derivative drugs have a molecular affinity for the peripheral nervous system that lacks a vascular barrier, leading to severe peripheral neurotoxicity that affects most cancer patients treated with cisplatin-based chemotherapy.

There are many studies that have shown that cisplatin induces peripheral neuropathy in rats or mice.Cisplatin successfully produced neuropathy in the CIPN models of primor studies.
