Systemic Lupus Erythematosus (SLE)
● Symptoms and Causes
SLE is a complex heterogeneous disease characterized by autoantibody production and immune complex deposition followed by damage to target tissues. It's what most people mean when they refer to "lupus". The most common symptoms include: skin rashes, pain or swelling in the joints (arthritis), swelling in the feet, and around the eyes (typically due to kidney involvement), extreme fatigue, low fevers.
Experts don’t know what causes systemic lupus erythematosus, but lupus and other autoimmune diseases do run in families.

●Pristane Induced C57BL/6 SLE Model 【Mechanism】Pristane is a mineral oil that was given intraperitoneally in C57BL/6 mice to induce peritoneal irritation and enhance the yield of monoclonal antibodies from ascites when hybridomas were subsequently injected. Satoh et al noted that pristane-injected mice, after a number of months, developed a lupus-like disease with immune complex glomerulonephritis, mild erosive arthritis and many lupus-associated autoantibodies. |
●TLR-7 Agonist Induced C57BL/6 SLE Model 【Mechanism】Several studies have demonstrated that altered Toll-like receptor (TLR) signaling contributes to the initiation and/or exacerbation of lupus in humans and in murine models. In recent years, it has become apparent that TLR-7 and TLR-9, which sense single-stranded RNA and unmethylated DNA, respectively, contribute to the development of autoimmune diseases such as rheumatoid arthritis, SLE, and psoriasis. Mice treated with the TLR7 agonist imiquimod (IMQ) significantly up-regulated leading to systemic autoimmune disease. |
●ALD-DNA Induced BALB/c SLE Model 【Mechanism】The main characteristic of SLE is the production of anti-double stranded DNA (anti-dsDNA) Abs. Immunization with activated lymphocyte-derived DNA (ALD-DNA) could induce a systemic lupus erythematosus-like syndrome in syngeneic mice, closely resembling human SLE pathogenesis. This model may help to elucidate the mechanism in volved in SLE development. |
